How does Signet of Capture work

I haven't tried it yet, but am curious to know. Do you have to wait until after the boss is done casting the required spell, or do you use the signet as soon as you see the spell under the Boss' name (that is while he's casting it)?

Also if you already have the spell in your skill list do you waste the Signet of Capture by activating it on a skill you already have?
