To help ease newer members into the Lore Forum community and give them a base to help them learn, I have decided to create a summary of information we have gathered from our research, what we still don't know, and what are just theories.
However, as I am not the All-Perfect All-Knowing Creature of the Lore, I'll leave it open for comments, and add/edit information as needed. I hope this addition will make the Lore Forum less imposing and to assist in bringing more Lorebodies to the discussions here.
Oh, and before I forget! Almost all of these will include spoilers, so if you hate those with a vengeance, play through the game first!
Well, to begin, I suppose it is only right to mention that, probably the first bit of information we gathered was the name of the regions of Tyria. It was, as far as I know, before the Lore Forum was around, that Eratimus managed to translate the maps of Tyria. Thus, he discovered the names of the regions of the, now, large continent of Tyria.
These were the beginnings of the Lore Forum and the community as itself. However, let's not get into detail over that. Probably the most notable research here has been done on the Mursaat, although we still do not have much in the way of information in relation them. Yet we did learn that the Mursaat are the Unseen Gods of the White Mantle, they do have an ability to manipulate souls as a weapon and to contain them. We have yet to prove in any way an ability to cloak themselves and their cities though, making it difficult to determine exact locations of their cities or settlements.
The soul manipulation doesn't end there, a most notable contribution by one of our fellow Lorebodies, Mularc Templare, managed to discover how our Titan friends work. At least for the most part, he theorized that the Titans were actually souls that took on a physical form from their environments. This was later proven as fact and elaborated on in-game and he actually has his own NPC dedicated to the discovery.
Not as substantial, of course, but we here at the Lore Forum, or at least a few of us, determined the location of Cantha before Factions was ever released. Well, we had a basic idea of it, from various NPC dialogue. It is still a commonly argued point though, as to the exact location. Some think it is north of Tyria, some think it is directly below Elona, but most of us at the Lore Forum believe it is south of Tyria and southwest of Elona.
Another topic, still being discussed today, is the mysterious Giganticus Lupicus and whether it was aquatic, land-dwelling, or amphibious. It is thought by some that it is aquatic and so far it seems the most likely of arguments. However it is still a topic to be debated.
An almost dead topic, that isn't discussed much today as it was in the past, is the connection between Ascalon and the Crystal Desert. It has been theorized that somehow Ascalonians made it into the Crystal Desert and managed to make settlements there, but as of yet there is no substantial evidence to back it up. The only pieces of evidence for this view, are the statue heads in the Catacombs.
That's all I can think of at the moment in terms of theories and discoveries on our part.
What we know very little about..
The top thing that comes to mind are the Enchanted Armors in the Crystal Desert, theorized to be Mursaat Armors enchanted by the Forgotten. However, we don't have enough evidence other than similarities in anatomy to back this up. One view, I think, is that they were prototypes to the Jade Armors we see today.
The Talmark Pillars! Probably one of the most mysterious structures in the game without any explanation or theory behind just what on Tyria it could be. We have no evidence of what civilization designed it, why it was made, or anything else.
The mysterious rocks in The Wilds. These have many views surrounding them with no solid theory to give them explanation. Some say they were made by the Centaurs living in the area. Some say they relate to the Flameseeker Prophecies. Some say they are related to the Dragons that awaken and are ravaging the land in GW2. We just don't know.
The Wizard's Tower of Kryta. A structure most everyone's probably seen once or twice if they helped people in the quest, the Villainy of Galrath. We do know there's a library in there that contains magical information and that Galrath was after it. We don't, however, know where on Tyria it came from or who designed it.
The Wizard's Folly Tower of Pre-Searing Ascalon. Another mysterious tower grounded in the mountains of Ascalon where an Elementalist is experimenting on a new spell. We do not know who built this tower or why, but we do notice that it has chains to pillars that have similar runes to the Talmark Pillars. Another mysterious thing is a "golem" that looks like an Enchanted Armor. How do these all connect? Well, I wish we had an explanation.
Majesty's Rest of Kryta. We know that the Scepter of Orr was found there, but why is Rotscale and an army of Undead there? We have a few theories floating around in relation to it, but we are still without a solid one to give a good explanation.
I'm forgetting something here..I know it.
Anyway, I hope this helps answer some questions some may have skimmed through tons of threads to get, and if I wasn't thorough enough I have links here at the bottom for assistance!

No, really, feedback is lovely. If there are good reactions I'll do as I said and make similar threads in the other areas of the Lore Forum.
